In Brussels, star striker Robert Lewandowski struck first for Poland inside the opening 30 minutes, but the tables were quickly turned as Belgium successfully bounced back from their loss to the Netherlands on Friday.
Before half-time Castagne - who appeared for 84 minutes of the game - played a key role in the Red Devils finding parity, his stabbed pass allowing Axel Witsel to equalise.
There was then no looking back in the second period, Kevin De Bruyne putting the home side ahead before Leandro Trossard of Brighton & Hove Albion scored twice to take the game away from Poland.
Tielemans was next to play his part, his assist in the 83rd minute on the money for Leander Dendoncker to score a fifth, with substitute Ikoma Lois Openda completing the scoring in added time at Stade Roi Baudouin.
The result and scoreline sees Belgium hop above the Poles on goal difference, second in their group on three points after two games, though still three points behind the Dutch leaders. Belgium next travel to Cardiff to take on Wales on Saturday (7:45pm kick-off BST)
For Wales, Foxes 'keeper Danny Ward was restored to the starting line-up as, buoyed from their qualification for the World Cup at the weekend, the Dragons returned to UEFA Nations League action with a tough test at home to the Netherlands.
Ward played the first 45 minutes at Cardiff City Stadium, replaced by Adam Davies at the interval, with Wales falling behind on 50 minutes when Teun Koopmeiners found the target.
In a dramatic finale it appeared that Robert Page's side had rescued a point, Rhys Norrington-Davies diverting home a 92nd-minute header, only for Burnley striker Wout Weghorst to score a header of his own 100 seconds later and secure the victory.
After two games in the group stage, Wales are yet to earn a point, with the Belgians next up at the weekend.
Elsewhere, there was disappointment for Thanawat Suengchitthawon, who appeared for Thailand Under-23s as they lost 1-0 against South Korea Under-23s.
The Leicester City Development Squad midfielder came on as a substitute in the 64th minute, replacing Sittichok Paso, but the side were unable to get back on level terms with Go Jae-Hyeon scoring the only goal of the game for the Taeguk Warriors.
