The Leicester City centre-back, who has helped his club collect six clean sheets in their last eight Premier League outings, played from start to finish for the Black Stars at ZSC Stadium in Abu Dhabi.
Southampton's Mohammed Salisu opened the scoring for Ghana on 70 minutes before, shortly afterwards, substitute and Bristol City star Antoine Semenyo added a second.
Amartey's caps tally now stands at 46 ahead of this winter's contest in Qatar. Ghana's World Cup campaign opens against Portugal next Thursday (4pm GMT kick-off).
