The Academy graduate missed Saturday’s clash against Crystal Palace with a knee complaint and will not feature during the quarter-final tie against Pep Guardiola’s side.
City will also assess whether striker Jamie Vardy (fitness) will be available for selection after making a return to action this weekend following a groin problem.
“New injuries, no,” confirmed manager Puel. “[Ben] Chilwell is not match fit and not available for this game. He is still not available.
“After [Daniel] Amartey, of course, we will see if Jamie [Vardy] needs a rest between games because it was his first game after injury.”
Puel confirmed centre-back Harry Maguire is fit after he took a knock early on at Selhurst Park before completing his first 90 minutes since late October.
Meanwhile, Northern Ireland international Jonny Evans is suspended after picking up yellow cards against Wolverhampton Wanderers and Southampton in the previous rounds.
“I think Harry [Maguire] is okay [after Crystal Palace],” added Puel. “Also, [Jonny] Evans is suspended.”
