The Foxes, three-time winners of the competition, will travel to south Yorkshire to face Chris Wilder's men during the week commencing Monday 21 August.
City have won 29, drawn 33 and lost 31 of their previous encounters with the Blades, who currently play in the Sky Bet Championship.
Andy King scored the only goal when the sides last met at Bramall Lane in the second tier six years ago. The Club's Development Squad also overcame United 5-4 on penalties last term, following a 0-0 draw in the Checkatrade Trophy.
The fixture date, as well as ticket prices and sales arrangements, will be announced in due course.
