The Foxes were 2-0 winners at Pirelli Stadium, thanks to goals from Kelechi Iheanacho and Wilfred Ndidi.
They will now face Rovers in the second round, with the game to be played at Prenton Park during the week commencing 28 August.
Tranmere were the side Leicester beat in the 2000 final of the competition - and Martin O'Neill, the Manager on that day at Wembley, was the man to draw the Sky Bet League 2 side's name out of the hat on TV.
