The young Foxes, who qualified from a group including Hull City, Harrogate Town and Grimsby Town, recently sealed a penalty shootout victory at Salford City.
Tranmere, on the other hand, booked their place in the next stage of the competition with a 2-1 home victory over Manchester City Under-21s earlier this week.
All matches in the third round of the Papa John's Trophy will be played during the week commencing 11 January, 2021.
