For the Stoke triumph, Brendan Rodgers was able to welcome back Ayoze Pérez and Jonny Evans after both Foxes had recovered from their recent injuries.
A lively start in Staffordshire saw Jack Butland deny James Maddison, while Kasper Schmeichel was forced into action on 15 minutes to make a save from James McClean.
Maintaining their pressure, City again went close through Pérez, who also tested Butland, before striker Jamie Vardy also saw an effort turned away by the Stoke stopper.
Despite the visiting side’s domination in the opening stages, Stoke were awarded a penalty when Evans fouled Benik Afobe, but Schmeichel did well to save Sam Vokes’ spot-kick.
Level at the interval, both sides traded blows as Vardy first fired straight at Butland before Danny Batth was inches away from connecting with a dangerous cross from the left flank.
City finally had their breakthrough on the hour mark. A brilliant cross from Ricardo Pereira found Marc Albrighton, who chested the ball down before slamming it into the far corner.
However, Stoke restored parity in fortuitous circumstances 10 minutes later, when former Fox Nick Powell’s cross dipped over Schmeichel and found the net.
Determined to add another win to their pre-season record, Maddison nearly put City back in front, only to see his 25-yard free-kick tipped wide by Butland.
The Foxes maintained their pressure, though, and with six minutes to play found their winner, as left-back Ben Chilwell found Tielemans, who swept home unmarked at the back post.
Over in Rotherham, meanwhile, another City side drew 2-2 at New York Stadium following a brace from Kelechi Iheanacho.

Youri Tielemans bagged Leicester City's winning goal at bet365 Stadium against Stoke City in pre-season.
Leicester City dominated proceedings in Staffordshire, with 67 per cent possession and 20 attempts on goal compared to Stoke’s 11.
They were unfortunate to concede when Nick Powell’s cross looped over Kasper Schmeichel, but deservedly regained their advantage on 84 minutes.
Belgium international midfielder Youri Tielemans was in the right place at the right time to convert Ben Chilwell’s cross after Marc Albrighton was earlier on target for the Foxes.
Who impressed?
Ricardo Pereira set up Marc Albrighton's opener at bet365 Stadium.
Matt Elliott told LCFC TV: “He was the main outlet in the first half for sure and it was a bit of a mismatch on that far side of the pitch up against Joe Allen.
"Pereira was having an absolute field day with no end results. He was hitting crosses in the box, but nothing was happening as a consequence.
"In the second half, that proved to be different. He was there for Albrighton’s goal, he assisted in that.
"He was getting forward more and more regularly. He’s got such a good engine.”
