Shannon O'Brien scored twice, as Janice Cayman, Saori Takarada, Deanne Rose and Aileen Whelan all found the net as Leicester made it two wins from two in the FA Cup, confirming their space in the final eight of the competition.
There were three changes to the Leicester side that were beaten 2-0 by Manchester City last weekend in the Barclays Women's Super League, with Janina Leitzig, Josie Green and Missy Goodwin all coming in to the starting XI.
It was a slow start from the hosts, who found themselves a goal behind when a dangerous free-kick from Birmingham was turned into her own net from Goodwin after an unfortunate deflection.
The first chance for Leicester came after 16 minutes when Yuka Momiki fired a first time effort from the edge of the box, but directed it straight at Lucy Thomas who got her body behind it to turn it away.
Shannon O'Brien scored twice during City's win at Pirelli Stadium.
As time went on, City's confidence grew and they were soon back level when a clever ball from Takarada fed Cayman, who picked out the bottom corner to take her tally to six for the season.
Not long after that, the hosts were in front through O'Brien, who picked up the ball on the left wing, cut inside and fired a powerful effort past Thomas with less than half-an-hour played.
Former Leicester forward Libby Smith then drew Birmingham back level, when she ran past the City defence on to a through ball and found the net from range past an on-rushing Leitzig, but a second goal from O'Brien gave Leicester the advantage heading into the break, with the winger looking fired up for the game.
The second period of play saw Leicester put the game out of sight, first of all when Takarada opened her account with the Club, directing Rantala's free-kick goalwards with a powerful header, before substitutes Rose and Whelan rounded off the scoring.
Saori Takarada headed home her first Leicester goal in the second half at Pirelli Stadium.
It took just a matter of seconds for the Canadian international to make her mark on the game, when she drove forward at the Birmingham defence, cut inside a sent an accurate effort into the bottom right-hand corner.
Inside the final 10 minutes, Whelan confirmed victory in Burton as she pounced on a loose ball in the box and volleyed a low attempt past the Blues' goalkeeper, making it double goalscoring figures for City in the competition this term.
The win at Pirelli Stadium books the Foxes into the quarter-finals of the FA Cup, with the all-important draw taking place on Monday evening, live on BBC's the One Show.
